About Revolution Medicines

Revolution Medicines works on cancer treatments still in clinical testing, focusing on the biological pathways that drive RAS-related tumors. It is developing a number of drug candidates aimed at these pathways, including compounds targeting SHP2, SOS1, and mTORC1 signaling, as well as inhibitors directed at specific mutated forms of RAS such as KRAS and NRAS variants. One of its earlier candidates has moved into Phase 1/2 trials covering solid tumors including gynecological and colorectal cancers.

The company builds its pipeline around several parallel drug programs rather than a single product, with each candidate targeting a different mechanism involved in RAS-driven cancer growth. It works with Sanofi on research and development of SHP2 inhibitors, a partnership that covers one of its clinical-stage compounds.

Revolution Medicines was founded in 2014 and is headquartered in Redwood City, California. Its shares are listed on the NASDAQ under the ticker RVMD.

What they said
Revolution Medicines' Q2 2026 earnings call highlighted transformative clinical progress, particularly in pancreatic cancer, continued advancement in non-small cell lung cancer (NSCLC) development programs, and strong balance sheet support to execute near-term regulatory and commercial plans. Management emphasized paradigm-changing Phase III RASolute 302 results for daraxonrasib in previously treated metastatic pancreatic cancer (published in NEJM and presented at ASCO), resulting in FDA acceptance of a full NDA and an expedited EMA Phase I review. The company expanded patient access through an FDA-cleared expanded access program (EAP) serving >2,000 cumulative patients, advanced multiple ongoing and planned registrational trials (RASolute 303–305, 309 in PDAC; RASolve 301, 307, 308 in NSCLC), and continues to build its mutant-selective and multi-selective RAS inhibitor portfolio (including elironrasib, zoldonrasib, RMC-5127, RM-055). Financially, RevMed ended Q2 with $3.9B in cash and investments (including proceeds from April financings and a $250M royalty tranche), while quarter-over-quarter operating spend increased substantially (R&D $395M vs $224M y/y; G&A $110M vs $41M y/y), producing a net loss of $644M. Commercial readiness for a potential PDAC launch in the U.S. was emphasized (commercial teams ~60 reps, patient support, field access, MSLs). Management framed a data-driven development strategy, remained cautious on regulatory timing/details for Europe and accelerated approvals, and was measured but confident in responses during Q&A.
Guidance
- FDA has accepted full NDA for daraxonrasib for previously treated metastatic pancreatic cancer (PDAC); EMA has initiated a phase review (expedited). - Plan to complete enrollment in RASolve 301 (NSCLC) in 2026; initial readout expected in 2027. - Global RASolute 303, 304, 305 ongoing and enrolling; RASolute 309 initiated. - Expect data update and development plan visibility for colorectal cancer (CRC) in Q4 2026. - Identify recommended Phase II dose for RMC-5127 in H2 2026; initial clinical data in 2027. - Initiate first-in-human study of RM-055 in Q4 2026. - Initiate RASolve 307 (elironrasib + SOC in first-line RAS G12C NSCLC) in Q4 2026; RASolve 308 initiated for zoldonrasib in first-line RAS G12D NSCLC. - Preparing for potential commercial launch in PDAC subject to regulatory approvals; commercial team staffed (~60 sales reps).
Risks flagged
- Significant cash burn and rapidly rising operating expenses (R&D and G&A), leading to a larger net loss. - Regulatory uncertainty (timing and scope of EMA review, potential questions from FDA on expanded datasets, and whether accelerated approvals will be granted). - Competitive landscape and potential need to compete with other KRAS-targeting agents (e.g., Roche/divarasib) which could affect trial control arms and market dynamics. - Crossover risk in frontline PDAC trials and impact on OS/PFS analyses if access to commercial daraxonrasib after approval is widespread. - Reliance on partner data (e.g., Tango PRMT5 data) to inform combination strategies; early partner data require expansion and longer follow-up for conviction. - Remaining committed capital under Royalty Pharma is milestone-dependent (up to $1.5B), not fully available unless milestones are met.
